Plant anatomy studies are important to understand the structure of internal tissues and their functions in supporting plant life and as a basis for taxonomic identification and classification. One of the problems in this field is the limited anatomical data on local species such as Plumbago auriculata, which has the potential as an ornamental and medicinal plant. This study aims to observe the anatomical structure of the roots, stems, leaves and pollen of P. auriculata using manual preparation methods (free hand section), maceration methods and acetolysis methods. This study is a qualitative descriptive study. Observations include the types of tissues that make up the roots, stems, leaves, stomata types, and stomata distribution. In addition, observations were made on the structure of tracheal fibers and pollen. Data were analyzed descriptively based on the morphological characteristics of the observed tissues. The results show that the roots have a tissue structure of epidermis, cortex, endodermis, pericycle, pith, xylem, and phloem; The stem contains spiral and annular trachea and long fibers with tapered ends, the stem has a tissue structure of epidermis, parenchyma, phloem, xylem, and vascular bundles, while the leaves show anisocytic stomata. This study provides an initial contribution to the documentation of the anatomical structure of local tropical plants and can be used as a reference for classification studies and learning of plant anatomy.
